產品介紹 This gene encodes a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. This member catalyzes the reduction of a number of aldehydes, including the aldehyde form of glucose, and is thereby implicated in the development of diabetic complications by catalyzing the reduction of glucose to sorbitol. Multiple pseudogenes have been identified for this gene. The nomenclature system used by the HUGO Gene Nomenclature Committee to define human aldo-keto reductase family members is known to differ from that used by the Mouse Genome Informatics database. 醛糖還原酶是多元醇通路的關鍵限速酶,當血糖在正常生理水平時,它并不激活,它對葡萄糖的親和力較低。在高血糖狀況下(如糖尿病),磷酸己糖激酶飽和, 醛糖還原酶激活,促使體內的葡萄糖轉化為山梨醇。然而,山梨醇脫氫酶(sorbitol dehydrogenase,SDH)的活力并未相應地成比例增加,山梨醇轉化為果糖的效率沒有提高,山梨醇本身由于極性強不易通過細胞膜,在細胞內會形成蓄積,使細胞膜的通透性發生改變,并使細胞中Na+-K+-ATP酶活性下降,造成肌醇喪失,導致細胞代謝與功能的損害。
因眼睛和神經細胞等組織內醛糖還原酶的含量較高,糖尿病病人體內高血糖的環境使這一通路很容易被打開,造成對相關組織的病理損害,如糖尿病、白內障、神經病變、腎臟病變、視網膜病變、動脈粥樣硬化等糖尿病并發癥DCC的發生。
